Liverpool-based River will create a 10-minute film showing a 3D virtual journey from Shanghai to Liverpool to promote the city at the Expo's Liverpool Pavilion.
The Wood Street company will also create a series of shorter interactive films and presentations to promote Merseyside and the wider North West. It won the deal after an international tendering process.
River expects to recruit another ten people in the next 12 months after the contract win. The company is also splanning to develop a new digital studio in Liverpool.
Liverpool is the only UK city with a dedicated presence at the Expo, which runs for six months from May 1.
Organisers expect to welcome around 1m people to the Liverpool stand out of an estimated 70m visitors to the Expo.
River is the third Liverpool creative company to be appointed to a key role on the Expo project, alongside PR firm Paver Smith and design company Uniform.
Liverpool Vision's director of investment and enterprise, Mike Taylor, said he was pleased local companies had won three key Expo contracts.
He said: "River Media has established itself as a force in international exhibition content, digital 3D production and digital signage content. This kind of experience will be essential if we are to create the kind of compelling content which will create a buzz at the Expo."
